Abstract

Abstract:Objective To establish a quantitative latex enhanced turbidimetric immunoassay for determination of plasma myeloperoxidase (MPO) concentration. Methods 7 anti-MPO monoclonal antibody were selected from 3 different companies, the optimum double-antibody combination was picked out by the method of each monoclonal antibody crosslink latex particle and one by one match. The suitable latex and reaction concentration were analyzed. Consequently, the repeatability, linearity, sensitivity, specificity and recovery rate of the assay were evaluated by testing a large number of samples. Results Optimum double-antibody was selected successfully. The quantitative MPO Latex enhanced turbidimetric immunoassay was set up by monoclonal antibody and the latex particles crosslink by chemical method. The results showed that the sensitivity: LoB=4ng/ml,LoD=6ng/ml,LoQ=12ng/ml, the within-run and interday- run coefficients of variation were:2.85% and 3.45%, and the mean recovery rate was 100.0%±1.2%, reference range was 54-266ng/ml. Conclusion An automated quantitative Latex enhanced turbidimetric immunoassay for determination of MPO is established. This method with high sensitivity, good stability, simply operation shows a better application to predict and diagnose cardiovascular diseases.
