Abstract:Objective To explore the effect of Lovastatin on BMP-2 during fracture healing process of mandibular fracture. Methods The fracture models of mandibular were established in with 24 rabbits. The rabbits were randomly and equally divided into medication group and control group, in which the rabbits were lavaged with Lovastatin of 10 mg/kg per day and placebo for 4 weeks since the next day after fracture. The expression of BMP-2 protein in bony callins were determined by PT-PCR and Western blot method on days 3,7,10,14,21,and 28 of fracture.The concentration of BMP-2 in serum was analyzed by ELISA. Results On days of 7 of fracture, BMP-2 gene expression was reach at the peak in medication group. However, the gene expression of BMP-2 in control group was delayed (on days of 14) in comparison with medication group. The results of HE stain showed that the positive cells of BMP-2 in experiment groups were higher than that in control group on days 7 and 14 (P <0.05), but on day 28 there was no different compared to control group (P>0.05). BMP-2 in serum were detected after fracture, and high expression of BMP-2 was detected on day 14 after fracture in medication group, which was higher than that in control group (P <0.05).Conclusion Lovastatin could promote the synthesis and secretion of BMP-2 during fracture healing, which can accelerate healing process of mandubular fracture.
