目的探讨红丝线草对二甲基亚硝胺诱导的肝纤维化大鼠肝组织TGF-β1及VEGF蛋白表达的影响。方法用二甲基亚硝胺诱导大鼠肝纤维化模型,不同浓度红丝线草干预后,取肝脏常规制片并HE染色,免疫组织化学方法检测各组大鼠肝组织TGF-β1及VEGF蛋白表达的情况。结果红丝线草高、中剂量组大鼠肝组织TGF-β1及VEGF蛋白表达较模型组明显降低(P<0.05)。结论红丝线草的抗肝纤维化作用,可能与降低TGF-β1及VEGF蛋白的表达而影响细胞外基质的代谢有关。
Objective To explore the effect of Peristrophe Roxburghiana on the expression of TGF-β1 and VEGF in hepatic fibrosis rats induced by dimethylnitrosamine . Methods The model of hepatic fibrosis rats induced by dimethylnitrosamine was intervened with different concentrations of Peristrophe Roxburghiana. The conventional flaking of liver and HE were used to observe histopathological staining change. The expressions of TGF-β1and VEGF in hepatic tissue were detected by immunohistochemistry. Results The expressions of TGF-β1and VEGF in hepatic fibrosis rats with high and middle dose of Peristrophe Roxburghiana were significantly lower than that of controls (P<0.05). Conclusion Peristrophe Roxburghiana may have a good curative effect of anti hepatic fibrosis. Its therapeutic mechanism may mainly be related to adjustment of metabolism of extracelluar matrix by decreasing TGF-β1and VEGF expression.
