摘要

摘要:目的 研究高迁移率族蛋白B-1(High mobility group box 1 protein,HMGB1)在心肌肥厚大鼠心肌中的表达变化及意义。方法 利用腹主动脉部分缩窄法在8~10周龄健康成年雄性Wistar大鼠体内构建压力超负荷心肌肥厚模型,并将大鼠随机分成两组:心肌肥厚组(OG)和假手术组(CG)。手术后按要求饲养4周后处死,观察离体心脏大小并计算心室重/体重比(mg/g)。取部分心肌标本,应用蛋白免疫印迹法(Western Blot)检测心肌组织中HMGB1表达水平。结果 与假手术组相比,心肌肥厚组心肌中HMGB1表达水平显著升高(P0.05)。结论 压力超负荷可引起心脏组织中HMGB1表达增加及活化,且增加的HMGB1可能参与心肌肥厚的发生和发展过程。

Abstract

Abstract: Objective To explore the expression changes and clinical value of high mobility group box1 Protein (HMGB1) in hypertrophic myocardium of hypertensive rats. Methods Pressure-overload models were built in healthy adult male wistar rats (8-10weeks old) by partial abdominal aorta ligature method. Wistar rats were divided randomly into 2 groups: the myocardial hypertrophic group and control group. After 4 weeks, they were killed. The in vitro cardiac size was observed and the ventricular weight/body weight ratio (mg/g) was calculated. Cardiac specimens were obtained. Westem Blot was performed to detect HMGB1 protein in myoeardium. Results Comparing with control group, the expression of HMGB1 Protein were increased significantly in myocardial hypertrophic group (P<0.05). Conclusion The pressure-overload may cause the expression of high mobility group box1 protein increase and activation. HMGB1 protein may be involved in the occurrence and development process of myocardial hypertrophy.
