手术前后翼状胬肉组织中内皮抑素、白介素-10的表达分析

摘要

摘要:目的 探讨手术前后翼状胬肉组织中内皮抑素(endostatin)、白介素-10(IL-10)的表达及其意义。方法 以我院收治的50例翼状胬肉患者为研究对象,手术切除的翼状胬肉组织为术前组标本。并在术后2周手术取得球结膜组织,收集标本为术后组。通过免疫组化法和Western blot 法检测两组标本中endostatin 蛋白的表达量,用ELISA法检测两组标本中IL-10浓度,并分别计算比较其阳性率。结果 免疫组化结果显示,术前组翼状胬肉组织中上皮细胞基底细胞层中的 endostatin抗原阳性表达明显少于术后组织。Western blot 检测结果显示,术前组endostatin 蛋白(0.60±0.12μg/L)显著低于术后组 (1.56±0.14μg/L), 术前组endostatin 蛋白阳性率(66.3%)显著低于术后组(92.5%),(F=5691.25,P<0.05), ELISA法计算得到术后组标本中IL-10的浓度(5.78±1.54ng/L)比术前组(9.51±1.81ng/L)显著降低,差异具有统计学意义(t=11.23,P<0.05)。术后组IL-10阳性率(54.6%)显著低于术前组阳性率(84.7%),(χ2=9.32,P<0.05)。结论 手术后翼状胬肉组织中内皮抑素表达水平随时间逐渐上升,白介素-10表达水平随时间逐渐下降。这两个指标的变化可以用来监测翼状胬肉手术的治疗效果、判断预后,同时可以用于术后监测复发。

Abstract

Abstract: Objective To investigate the expression and clinical significance of endostatin and IL-10 in pathogenesis of human pterygium before and after operation. Methods 50 pterygium patients were selected in this study. Excision specimens were collected as the pre-surgery group, and 2 weeks later conjunctival tissues were collected as the post-surgery group. Immunohistochemistry and Western blot were used to test the expression of endostatin. ELISA technique was used to test the IL-10 concentration. Results The expression of endostatin in the post-surgery group (1.56 ± 0.14μg/L) was significantly stronger than that of the pre-surgery group(0.60 ± 0.12μg/L),and the positive rate of endostatin in pre-surgery group was significantly lower than that of post-surgery group (P<0.05). The IL-10 levels in the post-surgery group(5.78±1.54ng/L)were significantly lower than that in the pre-surgery group (9.51±1.81ng/L) (P<0.05). The positive rates pre- and post-surgery were 54.6% and 84.7% respectively(P<0.05). Conclusion The expression of endostatin and IL-10 in pathogenesis of pterygium can be used as an indicator in monitoring effect of surgery, relapse and prognosis.
