目的 探讨可调微量移液器的操作对实验结果的影响。方法 使用血清、水、异戊醇和无水乙醇四种液体,从移液器量程的选择、改变吸头的填充方式、调节量程方式、吸头是否提前浸润、浸入时间、角度、深度和移液速度等几方面,分析影响移液准确性的因素,探讨可调微量移液器的操作方法。结果 可调微量移液器使用过程中,选择量程合适的移液器;移液器垂直对准吸头插入并左右小幅旋转填充吸头;调节量程需顺时针旋转刻度旋钮至超过量程的刻度,再回调至设定体积;提前浸润2~3次;浸入时间保持3s;吸头垂直(角度90度)浸入液面;浸入深度不宜过浅;慢速移液;以上操作能够显著提高移液准确性。结论 遵循良好的移液器操作规范对于得到准确的移液体积具有重要意义。
Objective To explore the effect of different pipetting manual on the accuracy of adjustable micropipette. Methods The adjustable micropipette with different ranges was used to deliver blood serum, water, isopropanol and anhydrous ethanol. The effect on the accuracy of pipetting was estimated at different conditions, such as the range adjusting, style of tip fixing, with or without tip pre-soaking, pre-soaking time, and angle of soaking, pipetting velocity and pipetting depth. Results The accuracy of adjustable micropipette was significantly increased when the target volume was close to the maximum range of pipette. In addition, vertical tip fixing, pre-soaking tip 2 to 3 times, deeply and perpendicular soak for about 3 seconds, slowly pipetting also increased the pipetting accuracy significantly. Most notably, that over-rotating the scale before setting to the target volume shows the best effect on improving the pipetting accuracy. Conclusion Good practice of adjustable micropipette could improve the experimental accuracy.
