云克对D-半乳糖致衰老小鼠血清自由基的影响

摘要

目的  探讨云克(99Tc-MDP对衰老模型小鼠胸腺、脾脏指数及老化相关酶的关系,对云克在抗衰老方面的研究及应用价值探讨有一定意义。方法  健康雌性昆明种小鼠48只,随机分为6组:正常空白对照组、模型组、维生素E组及云克低、中、高剂量组。正常空白对照组、模型组、维生素E组采用灌胃给药,云克三个剂量组采用腹腔注射给药,各组小鼠末次给药后,摘取眼球取血,而后颈椎脱臼处死,测定其血清SOD(超氧化物歧化酶)MDA(丙二醛)GSH-PX(谷胱甘肽过氧化物酶)含量,同时测定体重、胸腺重和脾脏重,求脏器指数。结果  与模型组小鼠相比,试验用药组小鼠胸腺、脾脏指数升高(P0.01P0.05);血清SODGSH-PX活性显著升高,MDA含量显著减少,差异有统计学意义(P0.01),云克组、维生素E组之间差异无统计学意义;模型组小鼠血清中SODGSH-PX含量显著减少,MAOMDA含量显著增加,与空白对照组比较差异有统计学意义(P0.01)结论  云克可能通过提高机体免疫功能,增加抗氧化酶的表达,减轻自由基对机体的损伤而起到延缓衰老的作用。

Abstract

Objective To investigate the effects of 99Tc-Methylenediphosphonate (99Tc-MDP) on serum antioxidant indexes and thymus and spleen index induced by D-galactose in aged mice. Methods Total 48 healthy female Kunming mice were randomly divided into 6 groups: normal control group, model group, vitamin E and low, middle and high dosage of 99Tc-MDP. After the last administration, the mice blood were took by removing the eye and the contents of malondialdehyde (MDA), activities of superoxide dismutase (SOD) and glutathione peroxidase (GSH-PX ) in serum were detected by biochemistry method. Meanwhile, the body weight, and the thymus and spleen of each mouse were measured. Results Compared with the model group, the thymus, spleen index in experimental treatment group were increased; the activity of SOD and GSH-PX in serum were enhanced significantly in the experimental treatment group, the serum MDA content were decreasedThe administration of 99Tc-MDP and vitamin E had no different degrees in the elevating SOD and GSH-PX activities, and reduction of the content of serum MDA. Compared with the normal control group, the activity of SOD and GSH-PX in serum were reduced significantly in the model group, the MDA content of serum were increasedConclusion 99Tc-MDP may reduce the effect of free radical damage to the body and play a role in delaying aging by increasing the immune function.
