补肾调肝清心方对围绝经期抑郁症睡眠障碍模型大鼠血清性激素以及IL-1β、IL-2、IL-6水平的影响

摘要

目的 观察补肾调肝清心方对围绝经期抑郁症睡眠障碍模型大鼠血清性激素以及IL-1β、IL-2、IL-6水平的影响。方法 将60只围绝经期大鼠按体重随机分为5组,每组12只:正常对照组(A);围绝经期抑郁症睡眠障碍模型组(B);围绝经期抑郁症睡眠障碍补佳乐治疗组(C);围绝经期抑郁症睡眠障碍米氮平治疗组(D);围绝经期抑郁症睡眠障碍中药治疗组(E);A组为围绝经期正常大鼠,其余4组进行18天不可预知慢性应激。于应激后对造模动物进行72h连续快速眼相睡眠剥夺(REM)。每天在应激前1h按组给药。在末次灌胃24h后,颈椎脱臼法处死大鼠,取静脉血,离心血清后,采用放免法测定大鼠血清中E2、LH、FSH含量以及ELISA检测大鼠血清中IL-1β、IL-2、IL-6含量。结果 围绝经期抑郁症睡眠障碍治疗组血清E2均高于正常组及模型组,且补佳乐及中药组与模型组相比有显著性差异(P<0.05),治疗组血清FSH均低于正常组及模型组,且米氮平及中药组与模型组相比有显著性差异(P<0.05),补佳乐与米氮平组血清LH均显著低于模型组(P<0.05);围绝经期大鼠在建立抑郁症睡眠障碍模型后IL-2、IL-6、IL-1β均显著低于正常对照组(P<0.05),补佳乐、米氮平及中药均能提高IL-2、IL-6水平,与模型组相比有显著性差异(P<0.05)。结论 模型组大鼠IL-2、IL-6、IL-1β水平降低;补肾调肝清心方可能是通过提高血清E2、IL2、IL-6水平及降低血清FSH、LH水平来治疗围绝经期抑郁症睡眠障碍。

Abstract

Objective To observe the effects of BushentiaoganQingxin decoction on sex hormone and serum IL-1 β, IL-2 and IL6 level in perimenopausal depression model rats. Methods 60 perimenopausal rats were randomly divided into 5 groups, including normal group, perimenopausal depression sleep disorder model group, perimenopausal depression sleep disorder progynova treatment group, perimenopausal depression sleep disorder mirtazapine treatment group, and perimenopausal depression sleep disorder TCM treatment group. After the last administration, the rats were killed by cervical dislocation. The serum levels of E2, LH, FSH, IL-1 β, IL-2 and IL-6 in each group rats were detected by RIA and ELISA. Results The serum E2 level in perimenopausal depression treatment group was higher than that of the normal group and the model group, and progynova and TCM medicine group compared with the model group(P<0.05). The FSH levels in treatment group was lower than that of the normal group and the model group, and mirtazapine and TCM medicine group compared with model group (P<0.05). The LH levels of the progynova and mirtazapine group were significantly lower than that of model group (P<0.05). The serum levels of IL-2, IL-6 and IL-1 β in perimenopause rats model of depression was significantly lower than that of normal control group (P<0.05). Progynova, mirtazapine and TCM could increase the serum IL-2 and IL-6 level compared with the model group (P<0.05). Conclusion The serum IL-2, IL-6 and IL-1 β levels in the rats model group was lower; BushentiaoganQingxin decoction may treat perimenopausal depression through increasing the serum E2, IL2 and IL-6 level and decreasing the serum FSH and LH level.
