Study on the Inhibitory Effect of Docetaxel on Mouse Melanoma with Lung Metastasis

Abstract

Abstract:Objective To investigate the inhibitory effect of docetaxel (Taxotere) on murine B16-F10 melanoma with lung metastasis. Methods The effect of docetaxel on cell growth and cell adhesion ability were detected by MTT method and cell adhesion experiment on mouse B16-F10 cells respectively. The mouse melanoma with pulmonary metastasis models were established with C58BL/6 mice by tail vein injection of B16-F10 cells and in vivo experiment were carried out to observe the effects of abdominal cavity injection of docetaxel on tumor metastasis of lung nodules in the lung tissue of mice. Results The in vitro cell studies showed that 20μg/mL of docetaxel had certain inhibitory effect on mouse B16-F10 cells growth and significantly reduced the adhesion ability of B16-F10 cells with matrix (P<0.05). The in vitro inhibition rate on B16-F10 cells growth with 5μg/mL, 10μg/mL and 20μg/mL of docetaxel at 24 h were 2.34%, 16.27% and 41.63% respectively. The tumor inhibition rate with abdominal cavity injection of 5μg/mL, 10μg/mL and 20μg/mL of docetaxel in mouse melanoma with pulmonary metastasis were 42.7%, 15.47% and 5.45% respectively. Conclusion Docetaxel could suppress the mouse melanoma with lung metastasis, and the mechanism may be related to its inhibitory effect on B16-F10 cell adhesion ability and growth ability.
